Most supplement brands talk about lion's mane the same way: "supports brain health." That's the safe sentence. It's also the lazy one.
Multiple clinical trials have published results in the last 18 months, and they paint a more interesting, and more honest picture than most companies want you to see. Some findings are genuinely promising. Others challenge assumptions the industry has been riding for years.
Here's what the science actually says right now.
What Lion's Mane Does at the Cellular Level
Lion's mane (Hericium erinaceus) contains two classes of bioactive compounds that matter for nervous system function: hericenones, found in the fruiting body, and erinacines, found in the mycelium.
Both have been shown in preclinical research to support the production of nerve growth factor (NGF) which is a protein your body uses to maintain, grow, and organize neurons. A January 2026 review in the International Journal of Molecular Sciences confirmed that these compounds enhance neurotrophin levels, particularly NGF and brain-derived neurotrophic factor (BDNF), and activate their associated receptor pathways.
In simpler terms: lion's mane contains compounds that may help your body do what it's already designed to do which is maintain and support healthy nerve cells.
That's a meaningful mechanism. But mechanism and outcome are two different conversations.
The 2025 Surrey Trial: Acute Effects in Healthy Adults
In April 2025, researchers at the University of Surrey published a double-blind, randomized, placebo-controlled crossover trial in Frontiers in Nutrition, one of the first to test acute (single-dose) cognitive effects in healthy younger adults (ages 18–35).
18 participants received a single 3g dose of a 10:1 lion's mane fruiting body extract (equivalent to 30g of fresh mushroom) or a matched placebo, with cognitive testing at baseline and 90 minutes post-dose. Seven-day washout between crossover arms.
What ImprovedPsychomotor performance. Participants showed significant improvements on the pegboard task for both dominant hand (p < 0.001) and non-dominant hand (p < 0.001), essentially fine motor speed and dexterity.
What Didn't ImproveGlobal cognitive composite scores and mood showed no significant effects. And notably, performance on executive function tasks (Flanker task, Trails B) actually declined in the active condition.
The honest takeaway: A single dose appears to affect motor processing but not the higher-order cognition most people associate with "brain supplements." The researchers themselves called the results inconclusive and emphasized the need for larger, longer trials.
The 2023 Northumbria Trial: Chronic Effects Over 28 Days
The Northumbria University pilot study (published November 2023 in Nutrients) tested 1,800mg daily over 28 days in 41 healthy adults aged 18–45.
Participants performed faster on the Stroop task (a measure of processing speed) after a single dose at 60 minutes (p = 0.005). After 28 days, researchers observed reduced subjective stress and an increase in circulating pro-BDNF—a precursor to the brain-derived neurotrophic factor involved in learning and memory.
Why This MattersThis is one of the first human studies to document a measurable change in a neurotrophic biomarker (pro-BDNF) following lion's mane supplementation. That's significant for the field, even if the sample size was small.
The Landmark Mori Trial: Older Adults with Cognitive Decline
The most frequently cited lion's mane trial remains Mori et al. (2009) which was a double-blind, placebo-controlled study of 30 Japanese adults aged 50–80 with mild cognitive impairment.
At 3,000mg daily over 16 weeks, participants showed significant cognitive improvement on the Revised Hasegawa Dementia Scale. Critically, those improvements began to reverse during the four-week washout period suggesting the effects require ongoing supplementation rather than being permanent changes.
This trial remains the strongest clinical evidence for lion's mane supporting cognitive function, but it's important to note the population: older adults with existing cognitive decline, not healthy younger adults looking for a performance edge.
What the 2025 Systematic Review Tells Us
A comprehensive systematic review published in Frontiers in Nutrition (Menon et al., 2025) analyzed the full body of evidence which included five randomized controlled trials, 15 laboratory studies, three pilot clinical trials, and additional study types.
The conclusion: lion's mane shows promising neuroprotective, antioxidant, and anti-inflammatory properties across study types. But the clinical evidence in humans remains limited by small sample sizes, short treatment durations, and the challenge that most studies are funded or designed differently, making direct comparison difficult.
The Dosing Question Nobody's Answering Well
Clinical trials have used doses ranging from 1,050mg to 3,000mg daily. Here's what that looks like in practice:

The optimal dose remains an open question. Extract type matters—fruiting body vs. mycelium vs. full-spectrum. Concentration ratio matters (a 10:1 extract at 3g represents 30g of fresh mushroom). And critically, the timing of measurement matters: animal pharmacokinetic studies suggest some active compounds peak at 8 hours post-administration, while most human trials test at 60–90 minutes.
We may be measuring too early to see the full picture.

The Bottom Line
Lion's mane is one of the most promising functional mushrooms in the research pipeline. The mechanism is sound. The early clinical data is encouraging. And the 2025–2026 wave of trials is starting to move the evidence from "traditional use" toward "clinical documentation."
But the honest position is that we're still early. The trials are small. The optimal dose, timing, and extract type haven't been established. And anyone telling you lion's mane is a proven cognitive enhancer in healthy adults is making a claim the science hasn't made yet.
That's not a reason to dismiss it. It's a reason to pay attention to who's being honest with you about what the research actually shows—and who's just riding the hype.
Every ingredient disclosed. Every dose intentional.
